He played as a running back and safety at Cathedral Preparatory School. Sanders had seven sacks in the high school state championship game. He played college ball at the University of Iowa where he forced three fumbles in his last game. Sanders was drafted in the second round by the Indianapolis Colts in the 2004 NFL Draft. He played for the Colts from 2004 until 2010.

He signed a five year $37.5 million deal in 2007 and became the highest paid safety in NFL history. In 2011 he signed a one year deal with the San Diego Chargers. His last four seasons on the NFL ended with Sanders being placed on IR. In 2005 and 2007 Sanders was selected to the Pro-Bowl and was a First-team All-Pro. He won a Super Bowl Championships with the Colts in 2007 and two AFC Championships. In 2007 Sanders was named AP NFL Defensive Player of the Year, GMC Defensive Player of the Year, AFC Defensive Player of the Year, and PFWA Defensive Player of the Year. He finished his career with over 300 tackles.
